The production of anti-human globulin serum (Coombs reagent) in goats.

چکیده
The rabbit is customarily used for the preparation of anti-human globulin serum, but there are several objections to the choice of this animal. These are: (1) The volume of serum obtained is small. To maintain supplies it is necessary either to have several animals producing the reagent, or to hyperimmunize and bleed the rabbits used too frequently, which in turn results in a less specific reagent. (2) The rabbit is prone to myxomatosis. This note reports the preparation of the reagent in the goat. The procedure has the following advantages over preparation in rabbits: (1) It yields larger volumes of serum. (2) The goat is not prone to infection with myxomatosis. (3) The immunizing and bleeding operations are simpler and quicker. (4) For the laboratory using or distributing large quantities of the reagent feeding and housing a goat is less costly than keeping 20 or 30 rabbits. Method

Extraction of a monospecific Coombs-reagent from chicken eggs.
During the last ten years the extraction of specific antibodies (ab) from the yolk of eggs of immunised chickens is more and more accepted as an useful alternative to the immunisation of mammals. The subject of this work is the immunisation of chickens with human IgG and the extraction of specific anti human IgG ab from egg yolk in order to obtain monospecific Coombs reagent. 12 Leghorn hens (2...
متن کاملDetection of Leukocyte Antibodies by Means of 1131-Labelled, Purified Anti-Human-Globulin Antibody. Problems of Nonspecific Adsorption of Globulin by Leukocytes
By ROBERT E. ANDERSON AND Ro L. WALFORD T HE PRESENT study forms part of a continuing effort to develop quantitative methods for detecting antigen-antibody reactions involving nucleated cells as antigen. Here we shall present information regarding the preparation of purified, 1131-labelled anti-human-globulin antibody (AHG antibody) by means of adsorption and elution from normal human globulin,...

متن کاملFactors affecting the speed of agglutination in the Coombs test.
The Coombs test (Coombs, Mourant, and Race, 1945a, 1945b, 1946) has become popular as a sensitive and reliable method of detecting incomplete antibodies. The essential reagent is the serum of an animal (usually a rabbit) which has been immunized with human globulin. Potent rabbit sera sometimes show a prozone phenomenon (Wootton, 1950; Van Loghem, Kresner, Coombs, and Fulton Roberts, 1950). The...

Background and objectives: Anti-human immunoglobulin G (IgG) antibody is produced against human IgG in various laboratory animals. The present study tried not only to produce anti-human IgG, but also to assess different antigen injection techniques leading to optimal production of anti-human globulin. Methods: The antibody was separated from human serum using precipitation method with sodium sul...
